P-postlabelling: a Sensitive Technique for the Detection of Dna Adducts

چکیده
Organisms including humans are exposed to many chemical carcinogens by both endogeneous and exogeneous sources. Of over 600 different agents evaluated for their overall carcinogenicity by the International Agency for Research on Cancer (IARC), more than fifty háve been considered to be carcinogenic in humans. Most of the chemical carcinogens are known to be activated to reactive intermediates that bind to nucleophilic centers in proteins and nucleic acids thereby forming covalent adducts'. The formation of DNA adducts is regarded as a critical early step in the multistage process of carcinogenesis. Moreover, approximately 90 % of the chemicals considered as carcinogenic for humans form DNA adducts. Although the majority of such DNA damage (DNA adducts) is eliminated by DNA repair processes, some persistent adducts often cause permanent mutations in important growth-controlling genes or loci, resulting in aberrant cellular growth and cancer". Therefore, DNA adducts represent a detectable and critical step in chemical carcinogenesis, and thus may serve as an early biomarker for cancer. Thus, the detection of the DNA adducts has become extremely important in risk assessment of chemicals as well as in determinations of exposures to carcinogens for humans. For an assay to be applicable in human exposure settings, it must (i) be sensitive enough to detect low levels of adducts; (ji) require only microgram quantities of DNA; (iii) give results quantitatively related to exposure; (ÍV) be applicable to unknown adducts that may be formed from complex mixture and (v) be able to resolve, quantitate and identify adducts.

Detection of platinum-DNA adducts by 32P-postlabelling.
We developed a sensitive 32P-postlabeling method for the detection of bifunctional intrastrand crosslinks d(Pt-GpG) and d(Pt-ApG) in DNA in vitro and in vivo. After enzymatic digestion of DNA the positively charged platinum adducts were purified from unplatinated products, using strong cation exchange chromatography. Subsequently the samples were deplatinated with cyanide, because platinated di...
متن کاملTamoxifen does not form detectable DNA adducts in white blood cells of breast cancer patients.
DNA from white blood cells of seven women receiving tamoxifen as adjuvant therapy for breast cancer and of three women who served as healthy controls was analysed for the presence of tamoxifen-DNA adducts using 32P-postlabelling with a limit of detection of 8 adducts/10(10) nucleotides. No postlabelled adducts with the chromatographic properties of known tamoxifen-DNA adducts were detected in a...
متن کاملDevelopment and validation of a direct sandwich chemiluminescence immunoassay for measuring DNA adducts of benzo[a]pyrene and other polycyclic aromatic hydrocarbons.
We have developed and validated a sandwich chemiluminescence immunoassay (SCIA) which measures polycyclic aromatic hydrocarbon (PAH)-DNA adducts combining high throughput and adequate sensitivity, appropriate for evaluation of adduct levels in human population studies. Fragmented DNA is incubated with rabbit antiserum elicited against DNA modified with r7,t8-dihydroxy-t-9,10-epoxy-7,8,9,10-tetr...
متن کاملAdduct formation on DNA and haemoglobin in mice intraperitoneally administered with styrene.
Styrene-specific N-7- and O6-guanine DNA adducts and N-terminal valine adducts were determined in mice tissues after the intraperitoneal administration of styrene. Blood, liver, lungs and spleen were collected 3 h after administration of various doses (from 0 to 4.35 mmol/kg b.w.) of styrene. DNA adducts were analysed by the modified 32P-postlabelling assay and N-terminal valine adducts were de...
متن کاملApplication of Rapid and Sensitive Real Time PCR Technique in Detection of DNA Impurities in Recombinant Interferon
Background & Objective: Interferon belongs to a family of cytokines, which has the most important role in the innate immune response to virus infections. While producing recombinant interferon in biological host, some pieces of host nucleic acids remain in product. Because of limitations in previous techniques for detection of these impurities, the objective of this study is to use rapid ...
